چکیده

A polarographic approach was developed for studying the biochemical physiology and biotechnological utility of the proteolytic anaerobe Clostridium sporogenes. Methyl viologen-mediated reductions were assessed in both analytical and preparative modes, using a dropping mercury electrode and a mercury pool electrode, respectively. Using these assays, we observed an active MV-linked NAD(P) reductase in toluene-permeabilised cells of this organism. Since this enzyme could be of use in the regeneration of pyridine nucleotide cofactors used in enzymatic biotransformations, the kinetics and stability of the enzymatic activity were determined. Reduced MV could also be oxidised by exogenous Dor L-proline in both intact and toluene-permeabilised cells, via the rudimentary electron transport chain and proline reductase enzyme possessed by this organism. The polarographic approach provided a novel, continuous assay for proline reductase activity. The proline reductase activity was greatest in the mid-to-late exponential phase of growth in batch culture. The reductive carboxylation of acetyl phosphate to yield pyruvate was also studied in toluene-permeabilised cells, and the coulombic yield calculated. Whilst acetyl phosphate was reduced stoichiometrically, pyruvate formation accounted for only some lo-15% of

Incidence of spore forming Clostridium sporogenes in different dairy products and their industrial and public health significance

Clostridium sporogenes is a food contaminating anaerobic bacteria causes late bowing effect in cheeses and spoilages in other dairy products. A total of 150 samples were studied for the presence of Clostridium sporogenes in raw milk, curd and cheese samples. Out of 150 sample 11(22%) Clostridium sporogenes was found in raw milk, 7(14%) in curd and 2(4%) in cheese samples. The study shows the ex...

First Complete Genome Sequence of Clostridium sporogenes DSM 795T, a Nontoxigenic Surrogate for Clostridium botulinum, Determined Using PacBio Single-Molecule Real-Time Technology

The first complete genome sequence of Clostridium sporogenes DSM 795(T), a nontoxigenic surrogate for Clostridium botulinum, was determined in a single contig using the PacBio single-molecule real-time technology. The genome (4,142,990 bp; G+C content, 27.98%) included 86 sets of >1,000-bp identical sequence pairs and 380 tandem repeats.

Genetic Characterization of the Exceptionally High Heat Resistance of the Non-toxic Surrogate Clostridium sporogenes PA 3679

Clostridium sporogenes PA 3679 is a non-toxic endospore former that is widely used as a surrogate for Clostridium botulinum by the food processing industry to validate thermal processing strategies. PA 3679 produces spores of exceptionally high heat resistance without botulinum neurotoxins, permitting the use of PA 3679 in inoculated pack studies while ensuring the safety of food processing fac...

Regulation of the arginine dihydrolase pathway in Clostridium sporogenes.

Arginine deiminase activity was induced during the vegetative growth of Clostridium sporogenes. The enzyme was sensitive to catabolite repression. The other enzymes of the arginine dihydrolase pathway, namely, ornithine carbamoyl-transferase and carbamate kinase, did not show such variation.
